about baclofen -derivative of gaba chlorophenyl derivative skeletal muscle relaxant antispastic agent and in trigeminal neuralgia. - -mechanism of action of baclofen -it inhibits both monosynaptic and polysynaptic reflexes at the spinal level by stimulating the gaba-b receptors which inhibits the release of glutamate and aspartate. it may also act at intraspinal sites producing cns depression. baclofen also exerts an antinoceptive effect - -indications for baclofen -1. spasticity in multiple sclerosis. -2. spasticity in spinal cord diseases (degenerative traumatic infectious or of unknown origin like amyotrophic lateral sclerosis) -3. transverse myelitis -4. spastic spinal paralysis. -5. muscle spasticity of cerebral origin e.g. cerebral palsy
